摘要
目的:合成和鉴定环丙沙星免疫抗原。方法:通过N-羟基琥珀酰亚胺活化酯法偶联免疫抗原。结果:经紫外光谱分析CIP-BSA偶联比(CIP:BSA)为6:1,紫外光谱峰形发生变化并位移。间接酶联免疫法分析证明其免疫兔所获得的抗体与环丙沙星有良好的特异性,半数抑制浓度为38 ng/ml。结论:成功合成了环丙沙星人工抗原,并获得了抗环丙沙星抗体,为ELISA或TRFIA等免疫方法的进一步研究提供了试验基础。
Objective:To synthesize and identify artificial antigen of ciprofloxacin.Methods:The immunogen CIP-BSA(ciprofloxacin-bovine serum albumin) were synthesized by the method of N-Hydroxysuccinimide active ester.These artificial antigens were identified by UV absorption method and the animal immunization test.Results:The coupling molar ratios of the hapten and carrier proteins were 6 for CIP-BSA.The antibodies showed the 50% inhibition of control values(IC50) of 38 ng/ml by ci-ELISA.Conclusion:These artificial antigens were successfully synthesized and the anti-ciprofloxacin antibody received,which provides the experimental basis for the further study of ELISA or TRFIA.
