氟伐他汀对诱导分化的HL-60细胞VEGF mRNA表达的抑制作用

摘要 目的探讨氟伐他汀(fluvastatin,Flu)对诱导分化的HL-60人早幼粒细胞白血病细胞(HL-60细胞)血管内皮生长因子(VEGF)mRNA表达的抑制作用及其机制。方法将体外培养的HL-60细胞随机分为空白对照组、阳性对照组、Flu处理组和Flu+甲羟戊酸(Mev)处理组。Flu处理组和Flu+Mev处理组加入终浓度为20μmol/L的Flu,Flu+Mev处理组在加入Flu的同时加入终浓度Mev100μmol/L,阳性对照组加入终浓度ATRA10μmol/L,空白对照组加入等体积二甲基亚砜(DMSO),作用72h后收获细胞。细胞处理72h后检测NBT还原能力;应用RT-PCR检测各组HL-60细胞VEGFmRNA表达水平。结果 Flu处理组细胞NBT还原能力(0.63±0.09)较对照组(0.22±0.06)显著升高(P<0.01),Flu+Mev处理组细胞NBT还原能力明显降低(0.34±0.04)(P<0.01),表明Flu已诱导HL-60细胞分化,而加入甲羟戊酸可以逆转Flu的诱导分化作用。阳性对照组和Flu处理组VEGFmRNA表达水平分别为1.51±0.02和1.58±0.04,均较空白对照组(1.87±0.05)明显下调(P<0.01);Flu+Mev处理组逆转由Flu诱导VEGFmRNA表达的下调(1.76±0.06),与Flu处理组比较具有统计学意义(P<0.05)。结论 Flu能抑制已分化的HL-60细胞VEGF基因表达,这种作用可能是通过"甲羟戊酸途径"介导的。
