摘要 目的研究Genistein与顺铂合用对肝癌细胞株HCCLM3的协同抑制作用.方法倒置显微镜观察Genistein与顺铂联用对肝癌细胞HCCLM3细胞形态的影响,MTT法检测两药联用对细胞增殖的影响,利用中效原理及公式计算联用是否具有协同作用;流式细胞术检测两药联用对细胞凋亡率和细胞周期的影响.结果倒置显微镜下Genistein与顺铂联用较单用组细胞密度降低,细胞相互分离,胞浆透亮度下降;Genistein与顺铂联用较单用组对HCCLM3细胞抑制率增加,Genistein与顺铂联用随着浓度增加对HCCLM3细胞增殖具有协同抑制作用;Genistein与顺铂联用较单用诱导细胞凋亡作用更明显,主要引起细胞G0/G1期阻滞.结论在体外Genistein与顺铂合用随着浓度增加对肝癌细胞能起到协同抑制作用;机制之一可能在于Genistein与顺铂联用较单用具有更强诱导肝癌细胞凋亡的能力. Objective To explore the synergistic effect of genistein and cisplatin on the proliferation of hepatoma cell line HCCLM3.Methods The inverted phase contrast microscope was used to observe the influence of combined cisplatin and genistein on the morphosis of HCCLM3 cells.MTT method was used to detect the effect of cisplatin combined with genistein on the proliferation of hepatoma cell line (HCCLM3 cells),and the median-effect principle and formula was used to computate the synergetic effect.The flow cytometry was used to detect the influence of the combined two drugs on cell apoptosis and cell cycle.Results By the inverted micro-scope,lower cell density was observed in genistein combined with cisplatin group than the single drug group. Cells in the combined drug group inter-separated,and the lucency of cytoplasm decreased.In genistein combined with cisplatin group,cell survival rate decreased more significantly than in single drug group.With the increase of drug concentration,the combination of genistein and cisplatin performed coordinate repression on the proliferation of HCCLM3 cells.Genistein combined with cisplatin had more significant effect in inducing apoptosis compared to the single drug,mainly by blocking the G0/G1 stage.Conclusions With the increase of concentration,genis-tein combined with cisplatin has synergistic repression on the proliferation of HCCLM3 cells in vitro. The mechanism may partly lies in that genistein combined with cisplatin possesses robuster capacity to induce apoptosis in cancer cells than they are single used.
