摘要
目的:研究成骨生长肽(Osteogenic growth peptide,OGP)对人牙周膜成纤维细胞(Periodontal ligament cells,PDLCs)在钛金属表面增殖分化的影响。方法:将纯钛试件放在12孔培养板内,取生长良好的第五代人牙周膜成纤维细胞接种在试件表面,加入不同浓度的OGP(10-11~10-7mol/L),分别在接种后1、3、5、7d应用MTT法检测细胞增殖;接种后3、7、10d应用酶联免疫法检测细胞内碱性磷酸酶(Alkaline phosphatase,ALP)活性的表达。结果:OGP在该浓度范围内时,对钛表面的人牙周膜成纤维细胞的增殖率有明显的促进作用(P<0.05),并且能促进细胞内ALP表达活性(P<0.05),最佳作用浓度为10-9mol/L。结论:OGP可促进钛片表面人牙周膜成纤维细胞的增殖活性,同时可以增强细胞碱性磷酸酶表达的活性,提示OGP在口腔种植领域中具有良好的应用前景。
Objective:To elucidate the effect of osteogenic growth peptide (OGP) on proliferation and differentiation of human periodontal ligament cells on titanium disc in vitro. Methods:Human periodontal ligament cells were cultured located on titanium disc individually in medium contained various concentrations of OGP;and cell proliferation quantity were examined by MTT and alkaline phosphatase activity were examined by enzyme-linked immunosorbentassay. Results:In vitro,the addition of OGP in cultures resulted in an increase of cell proliferation and ALP activity(P〈0.05). All OGP concentrations used (10^-11-10^-7mol/L)were effective but the maximum response was obtained with 10-9mol/L OGP. Conclusion:Our results demonstrate that OGP stimulates proliferation and ALP expression of human periodontal ligament cells located on titanium disc in vitro and suggest that OGP is a promising growth factor in the region of oral implant.
