HPLC法测定左金胶囊中4种生物碱的含量

Determination of Four Kinds of the Alkaloids in Zuojin Capsules by HPLC

目的:建立测定左金胶囊中4种生物碱含量的方法。方法:色谱柱为ODS柱(250 mm×4.6 mm,5μm);流动相为乙腈-0.05 mo.lL-1磷酸二氢钾溶液(52∶48,含25 mmol.L-1十二烷基磺酸钠,磷酸调至pH3.0);流速为1 mL.min-1;柱温为25℃;检测波长为225 nm,进样量为20μL。结果:盐酸小檗碱、盐酸巴马汀、吴茱萸碱和吴茱萸次碱的线性范围分别为12.18～121.8、3.016～30.16、0.081 55～0.815 5和0.042 96～0.429 6 mg.L-1,相关系数分别为0.9997、0.999 4、0.998 6和0.997 4;平均回收率分别为99.25%、101.80%、97.56%和96.19%,RSD分别为1.45%、1.39%、1.72%和1.87%(n=5)。结论:该法灵敏度高、重复性好,可用于左金制剂的质量控制。

Objective： To establish a HPLC method for the determination of four kinds of the alkaloids including berberine,palmatine,evodiamine and rutaecarpine in Zuojin capsules.Methods：HPLC system consisted of ODS column（250 mm×4.6 mm,5 μm）,mobile phase was acetonitrile-0.05 mol ·L-1 potassium phosphate which contained 25 mmol ·L-1 SDS（52 ∶48,adjusting pH to 3.0 with H3PO4）,the flow rate was 1 mL ·min-1,the column temperature was 25 ℃,the detection wavelength was 225 nm,and the injection volume was 20 μL.Results： It showed a good linear relationship between the peak areas and the concentrations of berberine,palmatine,evodiamine and rutaecarpine in the ranges of 12.18-121.8,3.016-30.16,0.081 55-0.815 5 and 0.042 96-0.429 6 mg ·L-1,respectively,and the correlation coefficients were 0.999 7,0.999 4,0.998 6 and 0.997 4,respectively.The recoveries were 99.25%,101.80%,97.56% and 96.19%,respectively,and RSD were 1.45%,1.39%,1.72% and 1.87%,respectively.Conclusion： The method is of high sensitivity and of good reproducibility.It can be used for quality control of Zuojin capsules.
