苯作业工人遗传损伤的研究

摘要 目的应用单细胞凝胶电泳法和胞质阻断微核法检测苯作业工人的遗传损伤,探讨检测苯作业工人遗传损伤和发生慢性苯中毒危险性的有效指标。方法选用各地已经确诊的慢性苯中毒患者40人为慢性苯中毒组,同单位的苯作业人员82人为对照组;检测两组人群单细胞凝胶电泳的彗星尾长、CB微核以及CB微核+3AB指数等指标;选用相关的统计学方法分析相关指标。结果两组之间CB微核率差异具有显著的统计学意义(t=3.033,P=0.003);此外,苯作业工人发生慢性苯中毒的危险性随3AB指数降低而升高(χ2趋势=9.100,P=0.003)。结论检测苯作业工人的遗传损伤应优选CB微核作为初筛指标,并且3AB指数在苯作业工人发生慢性苯中毒的危险性评价中有意义。 Objective Single Cell Gel Electrophoresis assay and Cytokinesis-block micronucleus method were used to measure the genetic damage in benzene workers to find the better index of detecting genetic damage and benzene poisoning risk in workers exposed to benzene.Methods 40 definite chronic benzene poisoning patients from various benzene plants were selected as chronic benzene poisoning group,82 benzene workers from the factories mentioned above were selected as control group.The comet tail length by single cell gel electrophoresis assay,CB micronucleus rate and CBMN+3AB index in the two groups were measured,then analyzed and compared the results using statistical methods.Results The results showed that there was some significant difference in CB micronucleus rates between benzene poisonings and their controls(t=3.033,P=0.003;additionally,the benzene poisoning risk was also increased with decreasing of 3AB index(χ2trend=9.100,P=0.003).Conclusions The results suggested that cytokinesis-block micronucleus method should be the better choice in evaluation of genetic damage of benzene workers,and 3AB index seems to be a valuable index in evaluating benzene poisoning risk of workers.
