砷酸钠(iAs^(5+))染毒大鼠尿液砷形态分析测定方法探讨 被引量:2

摘要 目的通过对染砷大鼠生物样品(尿液)中砷形态的分析,为进一步探讨砷代谢产物与砷毒作用机制间的关系提供基础数据,从而有助于进一步推测砷毒作用及其体内代谢机制。方法采用高效液相-氢化物发生原子荧光光谱法(HPLC-HGAFS)测定染砷大鼠尿液中砷形态的含量。结果尿样中iAS3+i、AS5+和DMA 3种物质的分离效果较好。该方法线性范围为20~100μg/L,尿样中iAS3+i、AS5+和DMA的相关系数分别为0.999 6、0.985 8和0.997 8。DMA的回收率为95.3%~108.4%,尿样中iAS3+i、AS5+和DMA的最低检出浓度分别为12.03、6.67和8.66μg/L。结论该方法具有良好的线性关系,灵敏度高,回收率高,结果准确。因此,该方法可以用于染砷大鼠尿样中砷形态代谢产物的分析。 Objective To analyze arsenic Speciation in biological specimen or urine of rat,and to understand further the basic data about relationship between it's metabolism and mechanism of toxicity in order to help conclude the metabolism and mechanism of toxicity in vivo.Methods The high efficiency liquid chromatography and hybrid genesis atomic fluorescence spectroscopy(HPLC-HGAFS) was applied to determine As3+ and it's product of metabolism.Results The isolation result of iAS3+,iAS5+ and DMA in urine were better.The linear range of method was 20~100 μg/L.The coefficient correlation of iAS3+,iAS5+ and DMA in urine were 0.999 6,0.985 8,0.997 8 respectively.The recovery rates of DMA was 95.3%~108.4%.The lowest detected concentration of iAS3+,iAS5+ and DMA in urine were 12.03,6.67,8.66 μg/L respectively.Conclusion The method is provided with favourable linear relation and better sensibility.Moreover,the recovery is fine and result is accurate.Thus,this method is suited for determination of the speciation of arsenic in urine.
