摘要
目的观察不同浓度二甲基亚砜(dimethyl sulfoxide,DMSO)对PC12细胞生长、形态与活力的影响,为DMSO作为脂溶性溶媒提供依据。方法用培养液将DMSO稀释到不同浓度,孵育细胞24 h后,采用显微镜观察细胞形态,四甲基偶氮唑蓝(MTT)显色法和流式细胞仪技术分别检测细胞活力和细胞凋亡。结果 DMSO终浓度低于2%时,PC12细胞能够正常生长并保持形态完整,细胞活力与对照组相比无差异,细胞未发生凋亡,但浓度超过4%时,细胞生长异常,细胞活力显著降低(P<0.01),细胞凋亡率梯度性增高。结论 DMSO对PC12细胞生长的影响存在剂量效应线性相关性,可作为细胞药物干预实验的脂溶性溶媒。
Objective To investigate the effect of dimethyl sulfoxide(DMSO) at different concentrations on PC12 cells growth,morphology,viability and provide a reference for using DMSO as a solvent mediator.Methods Different DMSO concentrations were dilluted in culture medium and incubated with cells for 24 h,cell morphology was observed under microscopy,cell viability and cellular apoptosis were tested by a colorimetric assay with 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide(MTT) and flow cytometry respectively.Results When the DMSO concentration was lower than 2%,PC12 cells were able to grow normally and keep intactly morphous,the cell viability was comparable to control group,no apoptosis was detectable,but at concentrations above 4%,cells became abnormal,cell viability decreased significantly(P0.01),cellular apoptosis was increased gradually.Conclusions The effect of DMSO on PC12 cell growth is linear and dose dependant.In the range of 0.01%~2%,DMSO can be used as a solvent mediator in drug treatment.
