植物金属硫蛋白提取液配方对镉中毒大鼠抗氧化作用的研究

摘要 目的研究新疆地产植物金属硫蛋白(MT)提取液配方对镉中毒的预防及治疗作用,为有效保护重金属作业人群,开发新型重金属中毒拮抗剂——金属硫蛋白提供理论依据。方法选择Wistar大鼠60只,分为阴性对照组,阳性中毒组,两个预防组及两个治疗组;用CdCl2进行染毒,采用前期试验所得到的植物金属硫蛋白提取液配方干预,预防组大鼠在镉中毒处理前两周以提取液进行干预,治疗组大鼠在镉中毒处理后两周以提取液进行干预。以血清超氧化物歧化酶(SOD)活力、谷胱甘肽过氧化物酶(GSH-Px)活力和丙二醛(MDA)含量为分析指标,分析MT提取液配方对镉中毒的治疗及预防效果。结果 (1)预防组,治疗组对血清SOD活力、GSH-Px活力、MDA含量的影响均与阳性对照组差异有统计学意义(P>0.05);(2)高剂量金属硫蛋白干预组MDA含量与治疗组差异有统计学意义(P<0.05);结论新疆地产植物金属硫蛋白提取液配方对镉中毒大鼠血清脂质过氧化损伤有一定的修复作用,且早期应用效果更好。 Objective To study the preventive and curative effects that extract of plant-metallothionein on cadmium poisoning rats,and to provide the theory basis for exploring a new type of heavy metal poisoning antagonist-metallothionein in order to protect the workers effectively.Methods 60 Wistar rats were used to establish the cadmium poisoning medols,rats were divided into six groups: negative-control group;positive-control group;two preventive groups and two treatment groups.Extract of plant-metallothionein intervented on preventive groups two weeks before CdCl-2 were used,and on treatment groups two weeks after CdCl-2 were used.We analyzed the SOD vitality,GSH-Px and MDA content in blood serum to inquiring the preventive and curative effects that extract of plant-metallothionein on cadmium poisoning rats.Results(1) There was significantly difference in activities of SDD,GSH-Px and MDA between positive-control group and preventive group or treatment group(P0.05);(2) The MDA content of high-dose preventic group was statistically significantly different from the therapeutic groups(P0.05).Conclusions The extract formula of metallothionein from Xinjiang local plants may have some reparation function on lipid peroxidation injury caused by cadmium poisoning,the effect was especially obvious in early stage.
