摘要
目的参照《中药材指纹图谱研究的技术要求(暂行)》,建立大蒜中氨基酸指纹图谱及技术参数。方法高效液相色谱法,以邻苯二甲醛为衍生化试剂,以0.045 mol/ml磷酸盐缓冲液∶乙腈∶1,4-二氧六环∶四氢呋喃(69.9∶25.0∶2.9∶2.2)为流动相,337 nm检测。10个不同产地大蒜所得的色谱图用《中药色谱指纹相似度评价系统(2004年A版)》分析评价。结果精密度、重现性、稳定性实验RSD不大于3%。指纹图谱共有峰6个,相似度在0.90以上。结论本方法稳定、准确,为进一步控制大蒜的质量提供依据。
Objective Determination of fingerprint and technical parameter of amino acids in garlic according to "Specification about fingerprint of Chinese herbal medicines(Tentative standard)".Methods A HPLC method was used with o-phthaldialdehyde derivatization,with the detection wavelength of 337 nm,the mobile phase was 0.045 M Phosphate buffer,acetonitrile,1,4-dioxane,tetrahydrofuran(69.9∶25.0∶2.9∶2.2).Ten samples from different habitats were analyzed and evaluated using the similarity evaluation system for chromatographic fingerprint of traditional Chinese medicine(Version 2004A).Results The relative standard deviation(RSD) of precision and reproducibility was not more than 3%.In the fingerprint of garlic,chromatographic peak owned by all of garlic samples was six.The similarity of samples was all above 0.90.Conclusion The method is accurate,convenient,stable and the chromatographic finger prints can be served as a powerful tool for the further quality control of garlic.
