摘要
目的:探讨miR-125a-5p和miR-125a-3p在胃癌中的表达情况及其与临床病理特征的关系。方法:采用茎环Real-time RT-PCR检测30例胃癌组织及其正常胃黏膜组织中miR-125a-5p和miR-125a-3p的表达并分析其表达与不同临床病理特征分组关系。结果:miR-125a-5p和miR-125a-3p在胃癌组织较癌旁正常组织表达均明显升高(P=0.012和P=0.025),其中miR-125a-5p表达量约为miR-125a-3p表达量的30倍,但二者表达比值在胃癌组织及正常胃黏膜组织中差异未见显著性;扩大样本量后结果显示miR-125a-3p的表达在有无淋巴结转移组间差异存在显著性(P=0.001);而miR-125a-5p的表达在不同分化程度组间差异存在显著性(P=0.003)。结论:正常胃黏膜及胃癌组织中均可以检测到miR-125a-5p和miR-125a-p的表达;miR-125a-5p和miR-125a-3p在胃癌组织和癌旁正常黏膜组织中的表达差异存在显著性,miR-125a-3p的表达在胃癌的不同淋巴结转移组间差异存在显著性;miR-125a-5p的表达在胃癌不同分化组间差异存在显著性。
Objective:To explore and discuss the expression of miR-125a-5p and miR-125a-3p and the correlation with clinicopathological factors of gastric cancer.Methods:The expression levels of miR-125a-5p and miR-125a-3p in 30 biopsies were examined using stem-loop real-time RT-PCR and the correlation between different clinicopathological factor groups were analyzed.Results:The expres-sion levels of miR-125a-5p and miR-125a-3p between tumor tissues and adjacent normal tissues were detected to be significant different in the 8 samples(P =0.012 and P =0.025,respectively).The expression level of miR-125a-5p was about 30-folded than that of miR-125a-3p while no significant difference of miR-125a-5p/miR-125a-3p value was detected between gastric cancer tissue and adjacent normal gastric tissue.The expression of miR-125a-3p was significantly different between different lymph node metastasis groups(P =0.001) and the expressions of miR-125a-5p was significantly different between cell differentiation groups(P =0.003).Conclusion:miR-125a-5p and miR-125a-3p can be detected in both gastric cancer tissue and adjacent normal gastric tissue.Expression levels of miR-125a-5p and miR-125a-3p have signicant difference between gastric cancer tissue and adjacent normal tissues.miR-125a-5p and miR-125a-3p are significantly expressed in different lymph node metastasis groups and cell differentiation groups,respectively.
