摘要
目的:探讨雷公藤内酯醇(TP)对阿尔茨海默病(AD)模型大鼠海马神经元突触后致密物中N-甲基-D-天门冬氨酸受体亚基2B(NR2B)和突触后致密物质95(PSD-95)表达的影响。方法:30只SD雄性大鼠随机分成对照组、模型组、用药组。应用免疫组织化学法和RT-PCR法检测各组大鼠海马NR2B和PSD-95蛋白和mRNA的表达。结果:免疫组织化学显色结果显示,用药组NR2B和PSD-95阳性细胞数较模型组均增多,平均光密度较模型组增加。RT-PCR结果显示,用药组NR2B和PSD-95mRNA水平较模型组升高。结论:TP能促进AD模型大鼠海马神经元NR2B和PSD-95的表达,表明其对突触可塑性有一定的改善作用。
Objective: To explore the effects of triptolide (TP) on the expressions of N-methyl-D-asparate receptor subunit 2B(NR2B) and postsynaptic density 95 (PSD-95) in the hippocampal neurons of rats with Alzheimer's disease (AID). Methods: Thirty male SD rats were equally divided into a control group, AD model group and TP-treated group at random. Immunohistoehemistry and RT-PCR were used to detect the proteins and mRNA expressions of neuronal NR2B and PSD-95 in the hippocampus respectively. Results: Immunohistochemical results demonstrated that the number of NR2B and PSD-95 positive cells in the TP-treated group was higher than that in the model group, and their average optical density significantly increased as compared with that in the model group. The rnRNA expression levels of NR2B and PSD-95 in the TP-treated group were significantly higher than those of the model group. Conclusion: TP can promote the expression of neuronal NR2B and PSD-95 in the hippocarnpus of model rats with AD, indicating that it could improve the synaptic plasticity of AID model rats.
