大鼠出生前后心肌端粒酶逆转录酶表达的变化

摘要 目的:探讨大鼠心肌组织端粒酶逆转录酶的发育规律。方法:免疫组织化学和免疫印迹技术检测SD大鼠心肌端粒酶逆转录酶(TERT)的表达,图像分析技术对心肌组织TERT的表达进行定量分析。结果:3组大鼠心肌组织中的心肌细胞、平滑肌和内皮细胞均有TERT的表达。TERT主要位于细胞质和细胞核内。TERT的总表达规律是20d胎鼠阳性表达强于出生后6d大鼠,出生后12d大鼠心肌表达最低,3组之间TERT的组间比较差异有统计学意义。结论:大鼠心肌组织的端粒酶活性随着日龄增加,其表达逐渐减少。 Objectives: To investigate the development regularity of telomerase reverse transcriptase (TERT) in myocardial tissue of rats. Methods: Immunohistochemistry and Western blot were used to detect TERT expression, and the quantity of TERT expression in the cardiac tissue was determined by computer image analysis. Results: The myocardial cells, smooth muscle cells and endothelial cells in the cardiac tissue of three groups all expressed the TERT. The TERT was located in cytoplasm and cell nucleus. The TERT expressed strongly at embryonic day 20, downregulated at postnatal day 6 and down to bottom at day 12 after birth. Significant differences were observed among the three groups. Conclusion: The expression of cardiac tissue TERT is decreasing with development of the heart.
