河豚毒素免疫抗原的制备及其表征体系的建立 被引量:3

摘要 为制备最适偶联率的河豚毒素(TTX)免疫抗原,并建立其表征体系,本研究采用非变性SDS-PAGE及紫外分光光度分析方法,探索TTX-甲醛-牛血清白蛋白(TTX-HCHO-BSA)免疫抗原合成体系中的偶联时间、偶联剂添加量及TTX与BSA质量比等反应条件对TTX免疫抗原偶联率的影响,并筛选出最佳的反应条件,即反应时间为72h、偶联剂添加量为12%、TTX与BSA质量比为1:5时TTX免疫抗原偶联率较大。同时,应用QuantityOne凝胶分析软件建立了BSA-TTX免疫抗原的偶联率计算方法,并探讨该免疫抗原的表征体系。 In order to prepare an effective tetrodotoxin immunogen with the optimal coupling rate, the effects of the coupling time, the amount of coupling agent formaldehyde (FA), and the mass ratio of tetrodotoxin (TTX) to bovine serum albumin (BSA) on the coupling rate in the immune antigen synthesis system of TTX-FA-BSA was studied by non-denaturing polyacrylamide gel electrophoresis and ultraviolet spectrophotometry analysis. The optimal reaction conditions were established using 72 h of coupling time, 12 % of coupling agent, 1:5 mass ratio of TTX to BSA. The calculation method of BSA-TTX immunogen coupling rate was established using gel analysis software Quantity One, and the characterization system for the immunogen was also discussed.
