黄芪甲苷乳膏对小鼠光老化皮肤的保护作用 被引量:6

Protective effect of topical use of astragaloside cream on photoaging skin in mice

摘要 目的探讨皮肤外用黄芪甲苷对紫外线(UV)照射引起的小鼠光老化皮肤的影响。方法 BALB/c小鼠84只,随机分成6组:模型组(A组)、模型+基质组(B组)、模型+黄芪甲苷低剂量组(C组)、模型+黄芪甲苷中剂量组(D组)、模型+黄芪甲苷高剂量组(E组)、正常对照组(F组),每组14只。模拟日光紫外线长期照射,形成皮肤光老化小鼠模型。C组、D组、E组照射同时外用黄芪甲苷乳膏。用组织切片HE染色、间苯二酚-碱性品红(Weigert)染色观察皮肤结构改变;以生化分析方法测定羟脯氨酸(HYP)含量。结果与F组比较,A组小鼠皮肤组织HYP含量明显降低(P<0.01),HE染色、Weigert染色病理切片呈现光老化状态;与A组比较,E组小鼠皮肤组织HYP含量升高明显(P<0.01),HE染色、Weigert染色病理切片显示抗光老化改变。结论黄芪甲苷乳膏对紫外线照射引起的小鼠皮肤光老化具有保护作用。 Objective To investigate the influence of topical use of astragaloside cream on the ultraviolet(UV)radiation-induced photo-aging skin in mice.Methods BALB/c mice were randomly divided into six groups of photoaging model(group A),model plus matrix(group B),model plus low dose astragaloside(group C),model plus middle dose astragaloside(group D),model plus high dose astragaloside(group E)and normal control(group F)with 14 mice each.The mouse model of photoaging skin was established by long-term UV radiation.Astragaloside cream was used on mouse back in groups of C,D and E before UV radiation.Histological sections stained with HE and Weigert were used for observing the structural changes in skin.HYP was detected by biochemical method.Results Compared with group F,HYP in group A decreased significantly(P0.01).Pathological changes of photoaging skin were seen under microscope.Compared with group A,HYP increased significantly(P0.01)and pathological changes of photoaging skin stained with HE and Weigert were improved apparently in group E.Conclusion Astragaloside cream has a protective effect against photoaging.
