三羟基异黄酮对ACC2及ACCM细胞株侵袭能力的影响

摘要 目的:观察4,5,7-三羟基异黄酮(Genistein)对腺样囊性癌细胞株(ACC2)及腺样囊性癌肺转移株(ACCM)的侵袭能力,和对两细胞株金属基质蛋白酶-2(MMP-2)蛋白表达的影响。方法:采用细胞划痕实验及Transwell小室侵袭实验,检测Genistein对ACC2和ACCM细胞侵袭能力的影响;采用明胶酶谱法及流式细胞术分别检测Genistein对两种细胞株MMP-2蛋白胞外分泌及胞内表达的影响。结果:Genistein能显著抑制ACC2及ACCM两种细胞株的侵袭能力(P<0.01),但两细胞株之间不存在差异(P>0.05)。同时,Genistein可使两细胞株细胞外分泌MMP-2蛋白的水平下调,及胞内MMP-2蛋白的表达降低(P<0.05)。结论:Genistein可对ACC2及ACCM的侵袭产生明显的抑制作用,该作用可能与诱导胞内外侵袭相关蛋白MMP-2的表达降低有关。 Objective: To study the effect of Genistein on the invasion of human adenoid cystic carcinoma ACC2 and ACCM cell lines in vitro. Methods: Effects of migration and invasion were assayed by the methods of transwell migration assay and scrape wound healing assay. The levels of MMP-2 in the ecto-medium were detected by gelatin zymography and gel imaging system. The MMP-2 level of the tumor cells was detected by flow cytometry. Results: The inhibitory effect on invasion occurred in the Genistein group was more potent compared with the control group (P〈0.01). Genistein could decrease the expression of MMP-2 by 29.81% and decrease the expression of activated MMP-2 by 17.63% in the ecto- medium of ACC2. It could also decrease the expression of MMP^2 by 41.25% and the expression of activated MMP-2 by 29.89% in ACCM. Genisitein significantly inhibited the expression of MMP-2 in tumor cells by the flow cytometry. Conclusion: Genistein significantly depresses the invasion of ACC2 and ACCM in vitro, and decreases the intracellular and extracellular MMP-2 expression.
