摘要
目的:本研究旨在检测瘦素、信号转导与转录激活子3(signal transducer and activator of transcription 3,STAT3)及其活化形式p-STAT3与其下游靶基因产物bcl-2在肺癌组织和癌旁正常肺组织中的表达情况,探讨瘦素、STAT3、p-STAT3和bcl-2表达之间的相关性及其与肺癌恶性程度及进展情况的关系。方法:采用免疫组织化学SP法检测52例肺癌及相应34例癌旁正常肺组织中瘦素、STAT3、p-STAT3及bcl-2蛋白的表达,并分析其与临床病理特征的关系。结果:瘦素、STAT3、p-STAT3及bcl-2的表达在肺癌组织中高于癌旁正常肺组织,且差异具有统计学意义(P<0.05)。STAT3和p-STAT3蛋白的表达与肺癌组织的临床分期和有无淋巴结转移有关(P<0.05)。在肺癌组织中,瘦素与STAT3、p-STAT3的蛋白表达水平之间呈正相关(P<0.05),p-STAT3与bcl-2的蛋白表达水平之间呈正相关(P<0.05)。结论:瘦素、STAT3及p-STAT3高表达可能与肺癌的发生发展有着密切关系,对其检测有助于判断肺癌的恶性程度及进展情况。
Objective:To detect the expressions of leptin, signal transducer and activator of transcription 3 (STAT3), p-STAT3, and bcl-2 in human non-small cell lung cancer (NSCLC) tissues and adjacent non-cancerous tissues, and explore the associations between the four proteins and their relationship with the malignant degree and progression of NSCLC. The relationships between the proteins and the malignant proceeding of NSCLC were also investigated. Methods:The immunohistochemical SP method was used to examine the protein expressions of leptin, STAT3, p-STAT3 and bcl-2 in 52 cases of NSCLC and 34 cases of para-cancerous normal lung tissues. Their correlations with clinical pathological features of lung cancer were statistically analyzed. Results:The positive expressions of leptin, STAT3, p-STAT3, and bcl-2 protein in NSCLC tissues were significantly higher than that in para-cancerous normal lung tissues (P〈0.05). The expressions of STAT3 and p-STAT3 had close relationships with TNM stage of lung cancer and lymph node metastasis (P〈0.05). The expression of leptin was positively correlated with the expressions of STAT3 and p-STAT3 (P〈0.05), and the expression of p-STAT3 was positively correlated with the expression of bcl-2 (P〈0.05).Conclusion:The high expressions of leptin, STAT3 and p-STAT3 may play an important role in the initiation and progression of human NSCLC. Detection of leptin, STAT3, and p-STAT3 is useful in the assessment of the malignant degree and development of lung cancer.
