肠易激综合征模型大鼠血清酪酪肽水平及其受体特性的测定 被引量:3

Determination of the serum level and receptors' characteristics of peptide YY in rats with irritable bowel syndrome

摘要 目的通过测定肠易激综合征(IBS)大鼠血清酪酪肽(PYY)水平及其空肠黏膜上皮细胞PYY受体的变化,探讨IBS的发病机制。方法将SD大鼠随机分成IBS腹泻型组(D-IBS组)、IBS便秘型组(C-IBS组)及对照组,应用同位素放射免疫法测定各组PYY水平及反映受体特性的两个参数:平衡解离常数(Kd)和最大结合容量值(Bmax)。结果①D-IBS组血清PYY水平为(66.50±32.10)pg/mL,与正常对照组的(15.33±10.63)pg/mL比较,差异有统计学意义(P<0.001);C-IBS组血清PYY水平为(19.51±17.62)pg/mL,与正常对照组比较差异无统计学意义(P>0.05)。②D-IBS组Kd=203.29±77.14,Bmax=645.71±184.30;C-IBS组Kd=347.43±162.76,Bmax=671.86±162.96,与正常组(Kd=328.75±92.11,Bmax=880.00±410.70)比较,差异均无统计学意义(P>0.05)。结论 IBS可能与血清PYY水平的高低有关而与其受体的变化无关。 Objective To explore the mechanism of irritable bowel syndrome by determining the serum level of peptide (PYY) in rats with (IBS) and changes of PYY receptors in jejunm epithelial cells in IBS rats. Methods SD rats were randomly divided into D-IBS group,C-IBS group and control group. We employed radioimmunoassay (RIA) method to measure the serum level of PYY in each group as well as the dissociation constant (Kd) and maximum binding capacity (Bmax),the two basic parameters reflecting the characteristics of PYY receptor. Results The serum level of PYY in D-IBS group (66.50±32.10)pg/mL was significantly higher than that in normal group (P0.001),but the serum level of PYY in C-IBS group (19.51±17.62)pg/mL did not significantly differ from that in normal group (P0.05). Kd and Bmax values in D-IBS group (Kd=203.29±77.14,Bmax=645.71±184.30) and C-IBS group (Kd=347.43±162.76,Bmax=671.86±162.96) were not significantly different from those in normal group (Kd=328.75±92.11,Bmax=880.00±410.70) (P0.05). Conclusion IBS may be related to the changes of the serum level of PYY,but not to the changes of PYY receptor.
