二球悬铃木叶片离体植株再生 被引量:3

摘要 研究不同浓度细胞分裂素BA或TDZ与生长素NAA或IBA组合对二球悬铃木种子苗所繁殖植株的叶片分化不定芽的影响.结果表明,诱导二球悬铃木种子苗所繁殖植株的叶片形成不定芽的最佳激素组合为3 mg/L BA和0.5 mg/L IBA;不定芽的诱导效果与植株的繁殖代数有关,在60 d的培养过程中,繁殖6代、10代、15代和20代的不定芽形成率分别为47%、44%、40%和38%.所有长达1 cm的不定芽在含2 mg/LIBA和1 g/L活性炭的培养基上培养20 d均生根良好.经过炼苗后,再生植株的移栽成活率达到92%. The effects of different combinations of BA or TDZ and NAA or IBA with different concentrations on adventitious bud induction from the leaves excised from the plants propagated from seedlings of Platanus acerifonia were investigated.The results showed that the combination of 3 mg/L BA and 0.5 mg/L IBA was the best one for the leaf explants to initiate adventitious buds;the bud induction rates were related to the propagation generations of the plants,and during 60 days of induction,47%,44%,40% and 38% of bud formation rates were obtained for the leaf explants of the plants propagated for 6,10,15 and 20 generations,respectively.All the buds up to 1 cm long rooted well in 20 days of culture on the medium containing 2 mg/L IBA and 1 g/L activated charcoal.After being acclimatized and then transplanted into soil,the regenerated plantlets grew well and the survival rate was up to 92%.
