摘要
目的:比较四种不同样品处理方法对人胎盘组织总RNA提取的影响,为RNA提取时的组织处理方法提供实验依据。方法:将新鲜人胎盘组织按0.1g的重量分成相等的80份,按照随机原则分成4组,每组分别采取不同的处理方法后置于液氮罐中保存并于日后用Trizol法进行总RNA提取,通过琼脂糖凝胶电泳和紫外分光光度计对RNA的浓度和纯度进行检测,所得数据进行统计学处理。结果:(1)提取成功率P<0.05,差异无显著性意义;(2)琼脂糖凝胶电泳总RNA样品较为完整,基本无降解;(3)纯度(OD260/OD280值代表)P<0.05,差异无显著性意义;(4)浓度P<0.05,差异无显著性意义。结论:尚无足够的证据证明,不同的处理方法对用Trizol法从人胎盘组织中提取总RNA有影响,在实际工作中我们可以采取较为简便、节约的方法。
Objective:To provide experimental evidence for extraction of total RNA from human placenta by handling the placenta differently.Method: Dividing the placenta into 80 shares by 0.1 gram per share,then dividing them into 4 groups randomly and handle each group with different methods separately.After that we preserve .them in liquid nitrogen,extract the total rna by Trizol and measure the RNA by agar gel electrosis and ultraviolet photometer.All of the datas are coped with statistics method.Result: (1)successful extraction rate, P〈0.05,the difference is of no significant meaning.(2)agar gel electrosis,the total RNA is intact and is not dissolved.(3)purity(OD260/OD280),P〈0.05,the difference is of no significant meaning.(4)concentration,P〈0.05,the difference is of no significant meaning.Conclusion:We have not had enough evidence to prove that the extraction of tatal RNA from human placenta is affected by different disposal methods.Thus,we can use the convenient and economic way to extract total RNA in daily experiment.
