摘要
目的:利用反相高效液相色谱法,研究益肝颗粒与淫羊藿提取物中淫羊藿苷在大鼠体内的药代动力学差别,探讨配伍对药效成分代谢的影响。方法:大鼠分别灌胃益肝颗粒、淫羊藿提取物后,于不同时间点尾静脉采血,通过建立的反相高效液相色谱及样品处理法,检测大鼠血浆中淫羊藿苷的浓度。采用药动学程序3P87对所得浓度结果进行统计处理,确定药动学模型及相关药代动力学参数。结果:益肝颗粒及淫羊藿中淫羊藿苷在大鼠体内呈二室开放模型,益肝颗粒与淫羊藿提取物在Tmax、Cmax、AUC、t1/2(a)、CL指标上比较,差异均有统计学意义。结论:复方对淫羊藿苷的体内过程有显著的影响,益肝颗粒中淫羊藿苷在大鼠体内的吸收增加,分布及达峰时间延长,血浆清除减少,生物利用度较淫羊藿药材高,产生的原因可能与复方中其他成分的相互作用有关。
Objective:Using high performance liquid chromatography to study the liver and particles and icariin in epimedium extract in rats Pharmacokinetics difference of compatibility of the active component metabolism.Methods:Rats were administered Yigan particles,epimedium extract,after tail vein blood at different time points,through the creation of high performance liquid chromatography and sample preparation methods,detection of icariin in rat plasma concentration.3P87 pharmacokinetic program using the obtained results were statistically treated concentration to determine the pharmacokinetic model and the relevant pharmacokinetic parameters.Results:Liver and particles and icariin in Epimedium in rats showed twocompartment open model,Yigan particles and epimedium extract Tmax,Cmax,AUC,t1/2(a),CL indicators comparison,the differences were statistically significant.Conclusion:The compound icariin in vivo process of the significant effects of icariin Yigan particles in rats increased the absorption,distribution,and peak time extension,reduction of plasma clearance,bioavailability than Epimedium herbal high,the cause may be related compound in the interaction of other components.
