摘要
目的观察雷公藤内酯醇对MRL/lpr狼疮鼠外周血清IL-2、IL-4、IFN-γ水平的影响。探讨雷公藤内酯醇治疗红斑狼疮的可能机制。方法选择30只MRL/lpr狼疮鼠和10只野生对照型MRL/MPJ小鼠,分为4组,狼疮鼠雷公藤内酯醇组、生理盐水组、未干预组、野生对照组,分别予以尾静脉注射雷公藤内酯醇(100μg/kg/d)和等量的生理盐水5d后24h后常规分离血清后,采用双抗体夹心法(ELISA)测定各组小鼠血清IL-2、IL-4、IFN-γ水平的变化。结果与野生对照型MRL/MPJ小鼠相比,MRL/lpr狼疮鼠血清中IFN-γ、IL-4含量升高(P<0.05,P<0.05),IL-2水平降低(P<0.05),经过雷公藤内酯醇治疗后,血清IFN-γ、IL-4水平降低(P<0.01,P<0.05),IL-2水平升高(P<0.01)。结论通过调节Th1/Th2细胞因子的失衡可能是雷公藤内酯醇治疗红斑狼疮的机制之一。
Objective To observe the effect of triptolide on the expression of IFN-γ、IL-2、IL-4 in MRL/lpr mice and to investigate the possible therapeutic mechanism of triptolide on lupus.Methods 30 MRL/lpr mice were randomized into triptolide treated group,normal sodium saline group and untreated group,and with 10 mice in each group.Each mouse of triptolide group was injected into the tail vein with 100ug/kg/d drug and each mouse of normal saline group with equal volume of saline one time per day for 5 days.To determine level of IL-2、IL-4 and IFN-γ in mice of each group were measured by ELISA.Results The expression of IL-4 and IFN-γ were significantly increased while IL-2 was significantly decreased in lupus mice compared with controls(P0.05,P0.05,P0.05).After the intervention of triptolide treatment,the levels of IL-4 and IFN-γ were decreased(P0.05,P0.01),but the secretion of IL-2 was increased(P0.01).Conclusion These results support the hypothesis that triptolide may induce a shift from Th2 to Th1 profile of cytokine secretion which might be one of the mechanism of triptolide in treatment of lupus.
