摘要
目的研究P-糖蛋白(P-glycoprotein,P-gp)在吗啡耐受大鼠脑内额叶皮层、海马及下丘脑表达差异。方法雄性SD大鼠12只随机分为吗啡耐受组和生理盐水对照组,吗啡耐受组连续皮下注射吗啡10mL/kg,2次/d,连续7d,建立吗啡耐受模型;生理盐水对照组同时按10mL/kg注射生理盐水。模型建立并确认成功后处死大鼠,取出大鼠海马、下丘脑和额叶皮层组织,以逆转录聚合酶链反应分析比较两组不同脑区P-gp基因表达。结果吗啡耐受组大鼠海马和下丘脑P-gp的表达显著高于同组的额叶皮层(P<0.05),并显著高于生理盐水对照组相同脑区P-gp的表达(P<0.05);而在生理盐水对照组各脑区间P-gp的表达差异无显著性(P>0.05)。结论海马和下丘脑P-gp表达增加明显高于额叶皮层,P-gp表达在吗啡耐受大鼠各脑区之间存在显著差异。
Objective To investigate the p-glycoprotein(P-gp)differential expressions in different brain regions of morphine tolerance rats.Methods Twelve male Sprague-Dawley rats were randomly assigned into two groups(n=6):morphine tolerance group and saline control group.Rats in morphine tolerance group were administered with morphine 10 mg/kg S.C,twice a day(8AM and 6PM)for 7 consecutive days while 10 mL/kg S.C of saline was given in saline control group.Morphine tolerance model was proved with anti-nociceptive effect examined with heat tail flick test.Rats were sacrificed for isolation of three brain regions(hippocampus,hypothalamus and frontal cortex).The expression of P-gp was detected by RT-PCR.Results In morphine tolerance rats,the P-gp mRNA expression were significantly higher in hippocampus and hypothalamus than in frontal cortex(P〈0.05)and was significantly higher than that in control group in the 3 brain regions.No significant differences of P-gp mRNA expression was revealed among 3 brain regions(P〉0.05)in control group.Conclusion P-gp mRNA expression in hippocampus and hypothalamus is higher than that in frontal cortex in morphine tolerance rats.
