摘要 取选优质的也门铁(Dracaena arborea)单株的顶芽和茎段作为快速繁殖的外植体,对各试验阶段所采用的培养基进行了对比分析,筛选出了比较适宜的培养基和比较有效的组培快速繁殖技术路线.结果表明,不定芽诱导培养基为MS+2.0 mg/L6BA+2.0 mg/LKT+2.0 mg/LAd+0.01 mg/LNAA,芽继代增殖培养基为MS+1.0 mg/L6BA+0.01 mg/L NAA+2.0 mg/L Ad,生根培养基为1/2MS+1.5 mg/L IBA时效果最佳;比较有效的组培快速繁殖技术路线为:无菌材料获取→不定芽诱导→壮芽继代增殖培养→炼苗与生根培养→试管苗移植和栽培管理. By taking the apical buds and stem as explants material from the superior single plant of Dracaena arborea,different media during each tissue culture stage was compared.It was indicated that the optimum media and technical step for tissue culture of Dracaena arborea were as follows:Adventitious bud inducing medium was MS+6 BA 2.0 mg/L+KT 2.0 mg/L+Ad 2.0 mg/L+NAA 0.01 mg/L;Bud proliferation medium was MS+6 BA 1.0 mg/L+ Ad 2.0 mg/L+ NAA 0.01 mg/L;Rooting medium was 1/2MS+ IBA 1.5 mg/L.The effective technical steps for tissue culture were Getting sterized explants→adventitious bud induction→bud proliferation→rooting and seedling growth training→transplantation and nusery growth of planlets in vitro.
