摘要 目的建立兔脓毒症模型。方法 18只新西兰兔随机等分为三组(A、B和C组)。A组和B组均在盲肠及阑尾交界处结扎,并分别用直径1.6mm和2.5mm针头对穿游离的阑尾3针,C组为假手术组。监测死亡时间和术前和术后12h两个时间点的白细胞、谷草转氨酶、肌酐、尿素氮、内毒素。对两个时间点检测结果的差值行多个样本均数的方差分析。对兔死亡后的肺脏行组织学检查。结果 B组动物的白细胞、谷草转氨酶、肌酐、尿素氮、内毒素变化与A组和C组之间差异均有统计学意义,A组和C组之间差异无统计学意义。B组动物肺脏在组织学检查上显示更明显的肺损伤,而A组和C组未见。结论在兔盲肠阑尾交界处结扎阑尾,用直径2.5mm钢针对穿阑尾3次可成功建立脓毒症模型。 Aim To prepare rabbit sepsis model. Methods There 12 New Zealand rabbits were randomly devided into A and B group .In all animals,sepsis was iduced by the cucum ligation an puncture model (CLP). CLP consisted of a dissection of the cecum and appendix,a ligation at the junction of cecum and appendix,and a triple punction of the isolated appendix with a needle that A group use the needle of 1.6 mm diameter and B group use 2.5mm . The survival time,levels of body endotoxin, creatinine(Cr),aspartate aminotransferase(AST),blood urea nitrogen(BUN),white blood cells(WBC)at pre-CLP and 12h after CLPweredetermined. Result Theplasmalevelsof endotoxin after CLP in B group animals was increased. The levels of Cr,AST and BUN were also increases(P0.05). Th count of WBCin in group B reduced. Conclusion Rabbit sepsis model was prepared by ligationing at the junction of cecum and appendix and puncturing appendix three times with a needle of 2.5mm diameter.
