转基因作物标记蛋白潜在致敏性的生物信息学预测 被引量:4

摘要 转基因作物表达的标记蛋白带来潜在致敏性顾虑。根据FAO/WHO决策方案,使用SDAP、Farrp和NCBI三大数据库对GUS、NptII和OPH的潜在致敏性进行评估。在线Blast或Fasta发现,OPH、GUS和NptII全序列或连续80aa的匹配性皆不存在致敏性可能。连续6aa比对中,OPH有2个片段与大豆、小麦和橡胶树的前纤维蛋白或德国蟑螂过敏原相关蛋白存在序列一致;GUS有1个片段与埃及伊蚊唾液腺过敏原存在序列一致;NptII有1个片段与摇蚊血红蛋白过敏原存在序列一致,但匹配的6aa序列皆未在各蛋白抗原性决定部位,是假阳性结果。初步证明转基因植物表达的GUS、NptII和OPH不具有潜在的致敏性风险。 Allergenicity prediction of transgenic expressed protein in crops is important. According to FAO/WHO recommendations,potential food allergenicity in GUS,NptII,and OPH was assessed in the databases of SDAP,Farrp,and NCBI,which were established for allergens prediction. Results of Blast or Fasta alignment online with full sequence or 80 aa window indicated that there was no allergenicity for sequence of GUS,NptII,and OPH. However,in OPH one 6 aa segment matched the sequence of profilin in soybeans,wheat or rubber tree,and another 6 aa segment matched the sequence of allergen-related protein in German cockroach. In GUS,one 6 aa segment matched the allergen "Aed a2" in salivary glands of Aedes aegypti. In NptII,one 6 aa segment matched the hemoglobin of Chironomus thummi. These identical segments did not show antigenicity,so it may be false positive for the matches.
