摘要 分别研究了摇瓶和发酵罐中二甲基亚砜对红霉素合成的影响。通过摇瓶实验确定了适宜的二甲基亚砜体积分数和添加时间,并在15 L发酵罐中考察了二甲基亚砜对红霉素合成代谢的影响。结果表明,在红色糖多孢菌发酵48 h时,向发酵罐的培养基中流加体积分数为0.16%的二甲基亚砜,红霉素效价可达7961 U.mL-1,比对照提高11.6%;二甲基亚砜可以提高羟基化酶——P450单加氧酶的含量,从而提高红霉素产量。 The biosynthesis of erythromycin by addition of dimethyl sulfoxide(DMSO) into medium in shake flask or in 15 L bioreactor were studied.The suitable volume fraction and adding time of DMSO were determined through shake flask experiment,and the effects of DMSO on the synthetic metabolism of erythromycin were explored in 15 L bioreactor.The result showed that,in the 15 L bioreactor,the chemical titre of erythromycin achieved 7961 U·mL-1which was increased by 11.6% of the control when 0.16%(volume fraction) DMSO was added into the medium at 48 h of fermentation of Saccharopolyspora erythraea.Adding DMSO could increase the content of P450,so that the yield of erythromycin was increased.
