摘要
目的研究新型复合材料3-羟基丁酸-co-3-羟基戊酸共聚物/溶胶-凝胶生物活性玻璃[poly(3-bydroxybutyrate-co-3-bydroxyvalerate)/sol-gelbioactiveglass,PHBV/SGBG]的体内成骨效能。方法制备犬胫骨骨缺损模型,实验组在骨缺损区植入PHBV/SGBG材料,对照组不作处理,分别于术后2、4、8、12周取材,大体标本及组织学切片观察该材料的体内成骨效能。结果实验组骨再生过程中,软骨成骨明显,术后2周骨缺损区软骨样组织较丰富;术后12周时,植入材料PHBV/SGBG基本完全降解,骨缺损区充填新生的成熟骨组织。对照组的骨修复不全,骨痂中可见纤维组织形成。结论复合材料PHBV/SGBG具有很好的骨传导性和骨再生能力,有望成为新型的骨组织工程材料。
Objective To study the osteogenesis efficiency of poly(3-bydroxybutyrate-co3-bydroxyvalerate)/sol-gel bioactive glass(PHBV/SGBG) composite in vivo.Methods Four bone defects were made in dog bilateral tibias.PHBV/SGBG composite were embed in the defects of the experimental group,while defects with no treatment served as control.Samples were harvested at week 2,4,8,and 12 post-surgery.The effect of osteogenesis was inspected by naked eye and histological observation.Results In the experimental group,endochondral bone was seen during the osteoanagenesis process.There was chondroid tissue formed in the bone defect within 2 weeks,and new mature bone complete healing the defects in 12 weeks while PHBV/SGBG composite almost resorbed at the same time.There was spontaneous ossification in the untreated group,but the healing was incomplete with fibrous tissue in the osteotylus.Conclusions PHBV/SGBG has good osteo-conductibility and bone regeneration capability.It may be a promising composite for bone tissue engineering.
