摘要
目的探讨单人建立稳定大鼠原位肝移植模型的手术难点及技巧。方法对200只成年雄性SD大鼠,采用改良的Kamada二袖套法,单人及双人裸眼下行大鼠原位肝移植各50对。结果单人与双人操作组供体手术时间、受体手术时间、无肝期分别为(27.22±1.03)min和(26.47±1.16)min、(50.58±3.10)min和(48.18±3.70)min、(11.60±0.11)min和(11.28±0.08)min。单人与双人操作组手术成功率、1周存活率、1个月存活率分别为90%和94%、88%和88%、84%和86%。结论改良的二袖套法,单人操作简便,成功率高,无需助手协助,可为大鼠原位肝移植实验提供稳定可靠的动物模型。
Objective To independently establish a stable rat model of orthotopic liver transplantation and discuss its operating difficulties and techniques.Methods A total of 200 adult male SD rats were adopted.The model was established with reformed two-cuff method by single operator and double operators.Fifty cases of rat orthotopic liver transplantations were performed with unaided eye each group.Results In single operator group and double operators group,donor operation time,receptor operation time and anhepatic time were(27.22±1.03)min vs(26.47±1.16)min,(50.58±3.10)min vs(48.18±3.70)min,(11.60±0.11)min vs(11.28±0.08)min.The successful rate,one-week survival rate,one-month survival rate were 90% vs 94%,88% vs 88%,84% vs 86% in single operator group and double operators group respectively.Conclusion The reformed two-cuff method is proved to be ideal for its advantages of simple operation,no assistant and high operative successful rate.
