日本血吸虫卵源性抗原诱导的抗肉芽肿病变保护性免疫调节机制的研究

摘要 日本血吸虫卵源性组分抗原SjR47可诱导小鼠产生明显的抗虫卵肉芽肿病变保护性免疫力。本文对免疫小鼠脾T淋巴细胞亚群表型动态和抗体免疫应答进行了研究,结果显示SjR47免疫小鼠的CD4+和CD8+T细胞水平,在感染后早期(4wk)升高,随后下降,至感染后8wk时,CD4+和CD8+T细胞比值<1.00,呈免疫抑制现象。SjR47免疫小鼠感染后0~12wk时血清抗SjR47抗体(IgG)水平显著高于抗SEA抗体水平,也显著高于同时期的佐剂和感染对照组小鼠抗SjR47抗体水平(P<0.01),进一步测定小鼠血清特异性抗体IgG及其亚类表明,SjR47免疫小鼠的IgG1水平升高非常显著。实验结果表明,SjR47免疫小鼠后诱导的抗虫卵肉芽肿病变保护性免疫力与细胞免疫及体液免疫均密切相关,其中IgG1抗体可能起着重要作用。 The protective immunity against egg granulomatous formation could be induced by SjR47 fractional antigen of Schistosoma japonicum eggs in BALB/c mice. In this paper, we studied on the kinetic changes of antibody and phenotype of splenic T lymphocytes in mice immunized with SjR47. The results showed that the levels of CD4 + and CD8 + T cells in immunized mice were high in early stage(4 wk) after infection and then decreased gradually, and the ratio of CD4 +/CD8 + T cells was smaller than 1.00 in 8 wk after infection. The anti SjR47 antibody(IgG) levels of sera from immunized mice were higher than those of anti SEA antibody in same mice and also higher than those of anti SjR47 antibody in mice of control( P <0.01). Further detection of IgG subclass showed the IgG 1 levels of immunized mice were significantly higher than those of control groups. These results indicated that the protective immunity of against granulomatous formation in mice immunized with SjR47 was related to cell and antibody immune and IgG 1 may play an important role im the against granulomatous formation.
