摘要
为了提高球形芽孢杆菌对中华按蚊的毒效,采用聚合酶链反应(PCR)技术成功扩增了B.s.中有毒效作用的基因片段,通过T4DNA聚合酶的修饰,使目的基因片段得以成功克隆。借助光敏生物素核酸标记探针,原位杂交筛选出目的基因阳性克隆。
Aiming at improving the toxicity of Bacillus sphaericus (B.s.) against Anopheles sinensis , we succeeded in amplifying the toxic gene fragment of B.s. with the help of PCR and getting its clone with the decoration of T 4DNA polymerase. The positive clones were screened by in situ hybridization with the probe which was labelled with biotin. In this study, T 4DNA polymerase played an important role in cloning the PCR products.
