摘要 目的探讨凋亡相关的半胱氨酸天门冬氨酸蛋白酶3(Caspase3)在蛋白酶体抑制剂MG-132诱导小鼠淋巴细胞白血病细胞株L1210细胞凋亡中活性的变化。方法分别以不同浓度的蛋白酶体抑制剂MG-132处理L1210细胞,流式细胞术检测细胞凋亡率,比色法测定细胞中Caspase3相对活性。结果在相同的作用时间下,随着MG-132浓度的增高(0、2.5、5、10、20μmol/L),细胞凋亡率及Caspase3的活性逐渐升高并存在剂量依赖性。结论 MG-132通过Caspase信号传导通路诱导L1210细胞凋亡并伴随Caspase3活性升高。 Objective To investigate the changes of Caspase3 activities in L1210 cell apoptosis induced by proteasome inhibitor MG-132. Methods L1210 cells were treated with different concentrations of MG-132 (0, 2.5, 5, 10, 20 μmol/L). Cell apoptosis and Caspase3 activities were detected by flow cytometry and chromometry, respectively. Results The apoptosis rates and the Caspase3 activities in L1210 cells increased in a dose-depended concentrations of MG-132. Conclusion MG-132 can induce apoptosis of L1210 cells through Caspase-transduced signal pathway by increasing of Caspase3 activities.
