活化后血小板对小鼠巨噬细胞分泌肿瘤坏死因子及凋亡的影响

摘要 目的初步探讨活化后血小板(PLT)对小鼠巨噬细胞分泌肿瘤坏死因子(TNF-α)及其凋亡的影响。方法体外培养小鼠巨噬细胞系RAW264.7细胞,用10mg/LLPS+20U/mlIFN-γ激活RAW264.7细胞,分别用1×104/ml、1×107/ml活化后PLT刺激被激活的RAW264.7细胞,TNF-αElisa试剂盒检测细胞培养上清中TNF-α水平,流式细胞术分析细胞凋亡情况。结果经活化后PLT刺激24h激活的RAW264.7细胞培养上清中TNF-α含量明显增加;活化后PLT刺激6d,凋亡明显被抑制。结论活化后PLT可促进小鼠巨噬细胞分泌TNF并抑制其凋亡。 Objective To explore the influence of activated platelets on apoptosis and the tumor necrosis factor (TNF-α) releasing of mouse macrophages. Methods Mouse macrophage cell line RAW264.7 cells were cultured in vitro, activated with 10 mg/L lipopolysaccharide (LPS) + 20 U/ml interferon-γ (IFN-γ). Then stimulate RAW264. 7 cells with 1 × 10^4/ml and 1× 10^7/ml activated platelets, detect TNF-α levels in cell culture supematant with TNF-α detection kit, flow cytometry analysis cell apoptosis. Results TNF-α contents in supematant significantly increased after 24 hours treatment of activated platelets, and apoptosis was inhibited significantly after 6 days treatment of activated platelets. Conclusion Activated platelets can promote secretion of tumor necrosis factor and inhibit apoptosis of mouse macrophage.
