摘要
目的:研究转化生长因子-β(TGF-β)与人类肿瘤增殖、侵袭和转移等的关系。方法:检索PubMed数据库,以"TGF-β、Smads和tumor"等为关键词,共检索到英文文献13964,选择以TGF-β/Smads信号通路在各类肿瘤发生、发展中的作用和意义为主题的文献作为主要参考。纳入标准:1)TGF-β信号转导和调节;2)Smads结构、功能及信号调节;3)TGF-β/Smads信号转导通入与肿瘤的发生、发展和转移的关系。根据纳入标准,最后纳入分析29篇文献。结果:TGF-β对肿瘤的作用是极其复杂的。在肿瘤发生初期,它通过诱导生长抑制,起到了肿瘤抑制子的作用,然而在肿瘤发生晚期,TGF-β通过促进肿瘤血管生成、抑制机体免疫功能而促进肿瘤细胞的浸润和转移。Smads是细胞内重要的TGF-β信号转导和调节分子,可以将TGF-β信号直接由细胞膜转导入细胞核内,当其功能发生异常时,将会影响TGF-β信号的传导,从而导致肿瘤的发生。结论:有关TGF-β/Smads信号通路的研究有可能为人类肿瘤的诊断和治疗方法提供新的科学依据,但在临床应用方面仍有很多问题需要解决。
OBJECTIVE:To explore the correlation between TGF-β and malignent biological behavior of human tumors.METHODS:A total of 13 964 articles were found in PubMed database,with TGF-β,Smads and tumor as key words.TGF-β/Smads signaling pathway in various types of cancer occurrence and the development of the role and significance of TGF-β/Smads signaling pathway in various types of cancers were chosn as the main reference of this review.Choice criteria:1)Signal transduction and regulation of TGF-β;2)Structure,function and signal conditioning.3)The relution between TGF-β/Smads signaling pathway and the ouurreme,development and metastasis of tumor.According to the above standards,29 were finally analyzed.RESULTS:It has been indicated that TGF-β is extremely complex on the role of tumor.In the early stage,it plays tumor suppressor which induces growth inhibition,however,TGF-β promotes tumor cell invasion and metastasis through promoting tumor angiogenesis and inhibiting immune function in the late stage.Smads are TGF-β signal transduction and regulation molecules of intracellular.TGF-β signaling can be directly transferred from the cell membrane into the nucleus by Smads,it will affect the TGF-β signal transduction and lead to tumor when its function is abnormal.CONCLUSION:TGF-β/Smads provides a new prognostic marker,but there are still many problems to be resolved in clinical applications.
