摘要
目的建立人工牛黄中胆红素的含量测定方法。方法采用Agilent TC-C18(250×4.6mm,5μm)色谱柱,甲醇-乙腈-2%冰醋酸溶液(72∶25∶3)为流动相;检测波长:452nm;流速:1.0mL.min-1;柱温:30℃。结果进样量在0.0272μg~0.2720μg范围内,呈良好的线性关系;r=0.9999(n=9),平均回收率为101.25%,RSD=1.66%,(n=9)。结论本法操作简便,准确,快速,可用于人工牛黄的质量控制。
OBJECTIVE To establish a method for the determination of bilirubin in cultured Calculus Boris.METHODS The separation was performed in the C18 column with the mobile phase of methanol-acetonitrile0.2% acetic acid glacical solution(72∶25∶3).The flow rate was 1.0mL·min-1 with the wavelength at 425nm,the temperature of column was 30℃.RESULTS The calibration curves were linear in the ranges of 0.0272μg~0.2720μg,r=0.9999(n=9),and the average recovery is 101.25%,RSD=1.66%,(n=9).CONCLUSION The method is quick,simple and accurrate,which can be used to control the quality of cultured Calculus Boris.
