摘要
目的研究替莫普利(temocapril)对大鼠损伤动脉血管平滑肌细胞(VSMC)增生的抑制作用和可能机制。方法测定大鼠动脉损伤后VSMC的增殖率变化;测定不同剂量、不同时期给予替莫普利对损伤动脉VSMC增殖率的影响;测定替莫普利对损伤动脉VSMC的血小板衍生生长因子(PDGF)-B和PDGF受体(R)β的影响。结果大鼠动脉损伤后2d和5d出现2个VSMC增殖期,并被10mg/(kg·d)替莫普利显著抑制。动脉损伤后,PDGF-B和PDGF受体β水平显著升高并被替莫普利抑制。结论大鼠损伤动脉VSMC的第1增殖期是由动脉损伤直接引起的,而第2增殖期只是第1增殖期的继发反应,是第1增殖期产生的PDGF作用于VSMC自身导致了第2增殖期的出现。替莫普利抑制VSMC增生的机制也是抑制PDGF及其受体。
[Objective]Study the inhibition of temocapril on the proliferation of the injured rat vascular smooth muscle cells (VSMC) and the mechanism[.Method]Some parameters were determined of the proliferation index alternation of VSMC of the injured rat artery,the proliferation index of the VSMC of the injured rat artery administrated with temocapril in different dose or period and the influence of temocapril on platelet derived growth factor (PDGF)-B and PDGF receptor (R)-β of the VSMC of the injured artery[.Result]The VSMC of the balloon catheter injured rat artery manifests 2 proliferation periods in the 2nd and the 5th day that were both inhibited by temocapril in 10 mg/(kg·d) drastically. PDGF-B and PDGF R-βlevel of the injured artery rose drastically and were inhibited by temocapril drastically[.Conclusion]The 1st proliferation period is caused by the injury directly and the 2nd one is only a derived reaction of the 1st one. It is the PDGF produced in the 1st proliferation period that causes the 2nd one, and the mechanism of the temocapril inhibiting the VSMC proliferation is inhibition of PDGF and PDGF-R.
