抑癌基因p16蛋白在肺癌组织中的表达及意义

Significance of p16 Protein Expression in Lung Cancer

目的：检测抑癌基因ｐ１６蛋白在７５例肺癌组织中的表达，并与癌旁组织和正常肺组织对比。结合肺癌的病理特征及预后进行分析，探讨ｐ１６基因与肺癌发生、发展的关系。方法：应用免疫组化ＳＡＢＣ法。结果：正常肺组织、癌旁组织和肺癌组织中ｐ１６蛋白阳性表达率分别为９１．７％、７５．０％和４９．３％，不同类型肺癌组织中ｐ１６蛋白的表达不同（Ｐ＜０．０５）；随着肿瘤分化程度的降低，ｐ１６蛋白的表达下降（Ｐ＜０．０５）；Ⅰ期和Ⅱ期肺癌组织中ｐ１６蛋白阳性表达率明显高于Ⅲ期和Ⅳ期（Ｐ＜０．０５）。生存３年以上肺癌ｐ１６蛋白阳性表达率明显高于生存３年以下者（Ｐ＜０．０５）。结论：ｐ１６基因缺失与肺癌发生有关，不同类型肺癌发生中ｐ１６基因的作用不同，ｐ１６蛋白表达缺失是肺癌的晚期表现，可作为判断肺癌生物学行为和预后的参考指标。

In order to study the significance of p16 gene in lung cancer, the expression of p16 protein in 75 cases of lung cancer, 12 adjacent tissues of cancer and 12 normal tissues were tested by immunohistochemical SABC method. The results showed as follows: the expression levels of p16 protein in the above three groups were 91.7%, 75.0% and 49.3% respectively. It was shown that the p16 protein expression significantly correlated with the type and grade of the tumor.The expression level was lower in poorly differentiated cancers than those in well differentiated tumors ( P <0.05), and it correlated with the clinical stage and survival time of the patients as well ( P <0.05). The results suggest that p16 gene protein plays an important role in carcinogenesis and progression of lung cancer and that it also correlates with the biological behavior and prognosis of lung cancer.
