摘要 目的:在成功克隆、表达和纯化人可溶性gp130(sgp130)分子的基础上,研究sgp130在白介素6(IL6)/白介素6受体(IL6R)系统中的生物学作用。方法:采用转人膜型gp130的BAF130细胞和人多发性骨髓瘤(MM)细胞株XG4CNTF和分离得到新鲜MM细胞,通过3HTdR的掺入做生物学检测,观察sgp130在上述细胞上对IL6生物学活性的影响。结果:sgp130对IL6和可溶性IL6R引起BAF130细胞的增殖有抑制作用。此外,sgp130对IL6刺激XG4CNTF细胞和分离得到新鲜MM细胞的增殖也有抑制作用。结论:sgp130为IL6的拮抗剂。 Objective:Study of the biological effect of soluble gp130(sgp130)molecule on the system of interlukin 6(IL 6)/IL 6 recepter on the basis of successful cloning,expression and purification of sgp130 molecule.Methods:The membrane gp130 transduced cell line BAF 130,human multiple myeloma(MM)cell line XG4 CNTF and freshly isolated MM cells were used for biological assa by means of 3HTdR incorporation in order to observe biological effect of sgp130 on IL 6 activity to these cells.Results:The results demonstrated that sgp130 displayed inhibitory role in the proliferation of BAF 130 cell line induced by IL 6 and soluble IL 6 recepter.In addition,sgp130 also showed the inhibitory effects on the proliferation,of XG4 CNTF cell lne stimulated by IL 6 and freshly isolated MM cells.Conclusion:These results suggested that sgp130 was an antagonist of IL 6.
