摘要
目的克隆并分析豚鼠BSEP基因全长cDNA序列,检测BSEP基因在胆结石豚鼠肝组织中的表达。方法采用3′、5′RACE方法扩增获得豚鼠肝组织BSEP基因全长cDNA,用生物信息学方法对其进行分析鉴定。实时荧光定量PCR检测BSEP基因在胆结石豚鼠肝组织的表达。结果豚鼠BSEP基因全长cDNA5 579 bp,共包含28个外显子,开放性阅读框(ORF)长为3 963 bp,编码蛋白长1 320个氨基酸。该基因在胆结石豚鼠肝组织的表达较正常豚鼠显著下调(P<0.01)。结论 BSEP基因在胆结石豚鼠肝组织中表达下调可能与结石形成有关。
Objective To clone and analyze the full-length cDNA sequence of guinea pigs bile salt export pump gene(BSEP) and to detect its expressions in cholelithiasis guinea pigs liver.Methods The BSEP gene full-length cDNA of guinea pigs was amplified by 3′and 5′RACE.Bioinformatic methods were applied to analyz the sequence.Real-time PCR was used to the quantification of BSEP gene expression in cholelithiasis guinea pig liver.ResultsThe length of the guinea pigs BSEP gene cDNA was 5 579 bp,containing 28 extrons and a 3 963 bp-length open reading frame(ORF) encoding a 1 320-amino-acid protein.BSEP mRNA expression in cholelithiasis guinea pigs liver was significantly decreased as compared to that of normal group(P0.01).ConclusionDown-regulation of BSEP mRNA expressions in cholelithiasis guinea pig liver may be involved in the formation of cholelithiasis.
