TNFα对嗜酸性粒细胞L-选择素表达的影响

EFFECT OF TNFα ON L SELECTIN EXPRESSION IN HUMAN PERIPHERAL EOSINOPHILS

多种细胞因子参与了哮喘时嗜酸性粒细胞与血管内皮细胞粘附的调节，但对嗜酸性粒细胞具有选择性作用的细胞因子尚未澄清。采用双色免疫荧光流式细胞技术，研究了炎前性细胞因子ＴＮＦα对人外周血嗜酸性粒细胞表面具有介导细胞粘附作用的粘附分子Ｌ－选择素和极迟抗原－４表达的影响，并与嗜中性粒细胞作以比较。ＴＮＦα对嗜酸性粒细胞和嗜中性粒细胞表面Ｌ－选择素均有上调表达的作用，而对极迟抗原－４表达无明显影响。实验结果说明，ＴＮＦα对Ｌ－选择素介导的嗜酸性粒细胞粘附无选择性作用。

Many cytokines participate in the interaction between eosinophils and vascular endothelium in asthma,but the selective cytokine has not been elucidated.The purpose of this study was to explore the effect of TNFα,an preinflammatory cytokine,on L selectin expression in human peripheral eosinophils and neutrophils by means of dual colar flow cytometry. The results showed that TNFα increase L selectin expression on both eosinophils and neutrophils,and had no effect on very late antigen 4(VLA 4,CD49d),indicating that TNFα has no selective effect on eosinophil adhesion mediated by L selectin.
