蛋白飞行质谱在恶性胸腔积液诊断的初步研究 被引量:2

摘要 目的用表面增强激光解析离子化飞行时间质谱(SELDI-TOF-MS)技术探寻恶性胸水蛋白标志物,建立快速、简便的恶性胸腔积液诊断方法系。方法收集恶性和非恶性胸水各50例,按照年龄、性别进行一一配对,采用WCX2磁珠技术对血清蛋白进行捕获,用蛋白质谱阅读器PBSⅡ对芯片进行扫描、分析。结果恶性和非恶性的胸水蛋白图谱相比,存在5个差异表达蛋白(P<0.05),其中包括3个高表达蛋白,分子量分别是5335、7577、11670Da。结论 SELDI-TOF-MS技术能筛选出灵敏度和特异性较好的差异蛋白,为恶性胸腔积液的早期诊断提供了一种新的实验方法 。 Objective To search specific protein biomarker for detecting malignant pleural effusion by SELDI-TOF-MS.Methods Thirty cases of pleural effusion and thirty controls were matched by gender and age.WCX2 protein chip was used to capture proteins,followed by PBSⅡ analysis.ResultsComparing to normal control,five protein biomarkers were identified which included three highly expressed proteins with the molecular weights as 5335,7577 and 11670Da,respectively.ConclusionsSome protein biomarkers with high sensitivity and specificity can be screened out by SELDI-TOF-MS technique.SELDI-TOF-MS provides a new method for early detection of malignant pleural effusion.
