摘要
利用分光光度法研究了光谱探针茜素红S(ARS)测定壳聚糖(CTS)含量的适宜条件,在pH5.0的NaAc-HAc缓冲液中ARS与CTS反应生成复合物,在530nm处产生新的吸收峰,反应体系在422nm和530nm处吸光值变化与壳聚糖含量呈线性关系,检测波长为422nm,线性方程y=0.6378-1.1334x(R=-0.997),在0~0.2mg/mL范围内呈现良好的线性关系,并成功地检测了样品康心胶囊中微量的壳聚糖。
Alizarin Red (AR) can interact with Chitosan(CTS) and the CTS-AR complexes are formed with the appearance of the new absorption peak at 530nm. The decrease in absorbance at 422nm and the increase in absorbance at 530nm are in proportion to CS concentration. Based on those above,a spectrophotometric method is proposed for the determination of CTS. The linear equation of CTS and absorption value at 422nm is :y=0.6378-1.1334x(R=-0.997). The linear range is 0~0.2mg/mL and the recovery range is 99.88%. Influence of a series of reagents on the absorbance of CTS-AR complex is also studied. The quantities of CTS in different compound samples can be determined in this way.
