摘要 目的研究前列腺增生上皮对间质细胞Latexin基因表达的影响。方法分离良性前列腺增生(BPH)的间质细胞与上皮细胞,经形态学及免疫组织化学方法证实后,构建共培养模型,并提取单独/共培养条件下间质细胞的mRNA,用RT-PCR检测Latexin基因表达情况。结果成功分离前列腺间质与上皮细胞并构建共培养模型,Latexin基因mRNA的表达在单独培养的间质细胞中表达,高于有上皮细胞共培养的间质细胞;Latexin/G3PDH的灰度比值单独培养的间质细胞为(1.122±0.30),有上皮细胞共培养的间质细胞为(0.917±0.24),差异有统计学意义(P<0.05)。结论前列腺增生上皮细胞可抑制间质细胞Latexin基因表达。 Objective To investigate the influence of epithelial cells on the Latexin expression of stromal cells in benign prostate hyperplasia(PBH).Methods The prostate epithelial cells and stromal cells were isolated,and then identified by the cell morphology and immunohistochemistry in the tissue of benign prostate hyperplasia.After the stromal-epithelial co-cultured model was constructed,the expression Latexin mRNA was detected by RT-PCR in stromal cells with or without the epithelial cells co-cultured.Results A stromal-epithelial co-cultured model was obtained.The expression Latexin mRNA was higher in stromal cells without epithelial cells co-cultured than that in stromal cells with epithelial cells co-cultured.The ratio of gray level between Latexin to G3PDH in stromal cells with or without epithelial cells co-cultured were(1.122±0.30)and(0.917±0.24)respectively(P0.05).Conclusion The epithelial cells can suppress the Latexin expression of stromal cells in BPH.
