摘要
目的探讨丹参作用类风湿关节炎患者成纤维样滑膜细胞bcl-2和bax基因mRNA水平的表达。方法关节液原代培养获取成纤维样滑膜细胞,分别用终浓度为0,0.195和0.39 g/L的丹参作用24 h后,提取总RNA并逆转录成cDNA保存,采用rt-PCR方法检测bcl-2和bax在mRNA水平的表达。结果以丹参浓度为0(bcl-2的相对表达量为0.55±0.27)作为溶剂对照组,bcl-2的表达在丹参浓度为0.195 g/L时(bcl-2的相对表达量为0.53±0.35)没有显著性差异(P=0.468),当丹参浓度达到0.39 g/L时,bcl-2的表达(相对表达量为0.25±0.17)显著性降低(P<0.05);bax的表达在2种丹参浓度条件下均无显著性差异(P均>0.05)。结论丹参可抑制类风湿关节炎成纤维样滑膜细胞bcl-2的表达,从而通过降低bcl-2/bax的值促进成纤维样滑膜细胞凋亡。
Objective It is to investigate the role of Danshen in patients with rheumatoid arthritis fibroblast-like synovial cells,and the expressions of bcl-2 and bax gene in mRNA level.Methods The primary culture of synovial fluid to obtain fibroblast-like synovial cells,and then treated with final concentrations of 0,0.195 and 0.39 g/L of Danshen.After 24 h,total RNA extraction and reverse transcription into cDNA,and using rt-PCR method to detect bcl-2 and bax expressions at the mRNA level.Results The concentration of Danshen was 0(bcl-2 relative expression levels of 0.55±0.27) as the solvent control group,bcl-2 expressions in the 0.195 g/L concentration of Danshen(bcl-2 relative expression levels of 0.53±0.35) were not significantly difference(P0.05),when the concentration of Danshen reach to 0.39 g/L,the bcl-2 expressions(relative expression levels of 0.25±0.17) were significantly lower(P0.05);the bax expressions in the two kinds of Danshen concentration were not significantly difference(P0.05).Conclusion Danshen have the ability of suppress the expressions of bcl-2 in the mRNA level of rheumatoid arthritis fibroblast-like synovial cells,thereby reducing the value of bcl-2/bax to induce fibroblast-like synovial cells apoptosis.
