摘要
研究重组人肿瘤坏死因子α衍生物A(rhTNFαDa)对离体造血细胞的抑制作用。方法骨髓细胞半固体培养和悬浮培养,DNA凝胶电泳,DNA缺口末端标记(TdT),电镜观察。结果在20ng/mlG-CSF存在下,rhTNFαDa能抑制小鼠造血祖细胞的集落形成,且rhTNFαDa对数剂量与形成的集落数呈负相关(r=-0.9845)。rhTNFαDa处理骨髓细胞48h,DNA凝胶电泳可见典型的阶梯状条带,TdT法可见大量的细胞核被染成棕色的骨髓细胞,电镜观察可见骨髓细胞核染色质变性、浓缩,细胞膜与核膜完整且明显皱缩。结论rhTNFαDa对造血细胞的抑制可能与其促进骨髓细胞凋亡有关。
Purpose To study the effects of rhTNFαDa on mice hematopoietic cells in vitro. Methods Semisolid culture,DNA agarose electrophoresis, in situ terminal deoxynucleotidyl transferase assays, view with electromicroscope. Results In colony assays,rhTNFαDa inhibited the growth of hematopoietic cells,and decreased colony formation,rhTNFαDa triggered apoptosis of total marrow,recognized by detection of apoptotic cells by DNA agarose electrophoresis, in situ terminal deoxynucleotidyl transferase assays and electromicroscope. Conclusions [WT10.BZ〗Apoptosis induced by rhTNFαDa may be responsible for its inhibitory effect on colony formation of hematopoietic cells.
