摘要
目的研究高原环境下大鼠运动疲劳后认知能力的改变及海马NR2B的表达。方法采用跑台运动方式建立大鼠重度运动疲劳模型,高原组大鼠由兰州直接引入到海拔4 767 m的可可西里高原进行疲劳运动;用Morris水迷宫记录大鼠的学习记忆能力;用免疫组化方法检测大鼠海马组织中NR2B的水平。结果大鼠海马NR2B的阳性表达位于细胞核,高原组较兰州组大鼠海马NR2B的阳性表达明显降低;与安静组相比,重度疲劳组的NR2B的阳性表达也有所降低。高原重度疲劳组、高原安静组及兰州疲劳组大鼠的空间学习记忆能力均受到不同程度的影响。结论高原环境和重度疲劳均可使大鼠的学习记忆能力受到影响,其机制可能与海马神经元中NR2B表达水平降低有关。
Objective To investigate the influence of exercise-induced fatigue on congnition and expression of hippocampus NR2B in environment of the plateau.Methods Severe exercise-induced fatigue models were established by treadmill exercise in rats.Plateau group were introduced from Lanzhou to Hoh Xil Plateau(4 767 m) to have fatigue exercise.The learning and memory ability of rats was recorded by Morris water maze,and NR2B levels in rat hippocampus detected by immunohistochemistry.Results The NR2B positive expression was located in the nucleus,and the hippocampus NR2B positive expression decreased in plateau group as compared with Lanzhou group.The NR2B positive expression of severe fatigue group was lower than that of quiet group.The spatial learning and memory abilities were affected to varying degreees in groups plateau severe fatigue,plateau quiet and Lanzhou fatigue.Conclusion High altitude and environment and severe fatigue can affect the learning and memory abilities of rats.Its mechanism may be related to the decrease of NR2B expression in the hippocampal neurons.
