组胺H4受体在变应性鼻炎大鼠鼻黏膜中的表达 被引量:3

摘要 目的观察新型组胺H4受体(H4R)在正常大鼠及变应性鼻炎模型大鼠鼻腔黏膜中的表达及分布,初步了解H4R在变应性鼻炎发病中的作用。方法 24只Wistar大鼠随机均分为模型组及对照组(n=12)。模型组采用卵清蛋白(OVA)法制备变应性鼻炎大鼠模型,对照组用生理盐水代替OVA,取两组大鼠鼻黏膜分别行免疫组化及RT-PCR检测H4R在蛋白质及mRNA转录水平的表达及分布情况,并进行对比。结果两组大鼠鼻黏膜非免疫细胞及免疫细胞的胞质及间质内均可见H4R表达,与对照组大鼠鼻黏膜中H4R蛋白和mRNA相对表达量(17656±5546,0.50±0.11)比较,模型组大鼠鼻黏膜中H4R的表达显著增加(28500±6575,0.85±0.15,P<0.01)。结论 H4R存在于正常大鼠鼻黏膜中,在变应性鼻炎模型大鼠鼻黏膜中表达显著增强,提示H4R可能是介导组胺参与变应性鼻炎发病的重要配体之一。 Objective To observe the expression and distribution of histamine H4 receptor (H4R) in nasal mucosa of normal rats and allergic rhinitis rats,and explore the role of H4R in allergic rhinitis.Methods Twenty-four Wistar rats were divided into two groups (12 each):normal control group (NC group) and allergic rhinitis model group (AR group).Allergic rhinitis model was reproduced by ovalbumin (OVA) method in AR group.In the control group (NC),OVA was replaced by normal saline.The nasal mucosa samples of rats in two groups were obtained,the expression and distribution of H4R at protein and transcription level respectively were detected by immunohistochemical method and RT-PCR,and the difference of expression between the two groups was then compared.Results Compared with the expression of H4R at proteins and transcription level in the nasal mucosa of rats in NC group (17656±5546,0.50±0.11),the expression of H4R increased significantly in the rats of AR group (28500±6575,0.85±0.15,P〈0.01).H4R was located at cytoplasm and intercellular substance area of non-immune cells as well as immune cells.Conclusions H4R exists in normal rats' nasal mucosa,the expression of H4R is enhanced significantly in allergic rhinitis rats.H4R may be one of the ligands which mediate histamine to participate in pathogenesis of allergic rhinitis.
