摘要
目的和方法:对大鼠肺微血管内皮细胞(PMVEC)培养、鉴定。观察了低浓度H2O2(0~100μM)对体外培养的PMVEC的迟发性损伤效应。结果:MTT法发现H2O2对PMVEC的增殖代谢活性具有剂量依赖性抑制作用,而且当H2O2去除后,此作用仍可进一步发展。形态观察、流式细胞仪DNA测定、3’末端标记DNA降解片断原位检测(TUNEL)均表明,细胞凋亡是H2O2对PMVEC迟发性损伤效应的一种重要形式。
AIM and METHOD:Pulmonary microvascular endothelial cells (PMVEC) were cultured and characterized.Delayed injury effects of low concentration of H 2O 2(0~100 μmol/L)on the PMVECs were observed.RESULTS:It was found with MTT method that H 2O 2 inhibited the proliferative and metabolic activity of PMVEC in vitro in a dose-dependent way,and after the removal of H 2O 2,this effect could develop further.Morphological observation,flow cytometry and DNA fragment in situ detection (TUNEL) revealed that apoptosis is an important form of delayed injury effect of H 2O 2 on the endothelial cells.CONCLUSION:Low concentration of H 2O 2 could induce a delayed effect on PMVEC: apoptosis.
