SHR血浆NPY、主动脉平滑肌细胞NPY受体的变化及药物的干预作用 被引量:1

摘要 目的:观察自发性高血压大鼠(SHR)血浆神经肽Y(NPY)、血管平滑肌细胞(VSMC)NPY受体的变化及美托洛尔、培哚普利干预的影响。方法:用放射免疫分析法和受体放射配体结合分析法测定血浆NPY及VSMC上NPY受体密度的变化。结果:SHR血浆NPY含量明显高于WKY,血压的高低与血浆NPY的量呈明显的正相关。培哚普利降压的同时亦降低SHR血浆NPY的量,而美托洛尔对NPY量则无影响;培哚普利引起NPY受体密度上调,而美托洛尔则使NPY受体下调。结论:NPY在高血压的发生、发展中可能起到重要的作用;在培哚普利的降压作用中可能有血浆NPY含量下降这一因素参与,美托洛尔致NPY受体密度下调可能在预防主动脉管壁结构重建中发挥着一定的作用。 AIM:To investigate the changes of plasma neuropeptide Y (NPY) levels, the NPY receptors on aortic vascular smooth muscle cells and the effects of perindopril,metoprolol upon spontaneuosly hypertensive rats(SHR).METHODS:NPY plasma levels and receptors were detected by radioimmunoassay and radioligand binding assay.RESULTS:Plasma NPY levels were significantly higher in SHR than that in Wistar-Kyoto rats(WKY);There was a close relationship between the height of blood pressure and plasma NPY levels.Perindopril could markedly inhibit release of NPY through the prejunctional facilitary angiotensin Ⅱ receptors,while the metoprolol could not.Perindopril induced upregulation of NPY receptor density;metoprolol induced downregulation of NPY receptor density.CONCLUSIONS:NPY might be involved in the pathogenesis of hypertension,the decrease of NPY levels might be involved in the antihypertensive mechanism of perindopril.Metoprolol induced downregulation of NPY receptor density,which might play an important role in the prevention of hypertrophy of vascular smooth muscle cells.
