摘要
目的:探讨L-精氨酸(L-Arg)对缺氧性肺动脉高压(HPH)大鼠血浆内皮素-1(ET-1)释放的影响。方法:将Wistar大鼠40只分为:对照组,缺氧组,缺氧+Nω-硝基-L-精氨酸甲脂(L-NAME)组和缺氧L-Arg组。结果:缺氧组的肺动脉平均压(mPAP)显著高于对照组(P<0.05),缺氧组+L-Arg组的mPAP显著低于缺氧组(P<0.05)及缺氧+L-NAME组(P<001),缺氧组的RV/(LV+S)比值显著高于对照组(P<001),缺氧+L-NAME组的RV/(LV+S)比值显著高于缺氧组(P<005)及缺氧+L-Arg组(P<001),缺氧组和缺氧+L-NAME组ET-1含量明显高于对照组和缺氧+L-Arg组(P均<001),mPAP与血浆ET-1含量呈明显正相关(r=0682,P<005)。结论:L-Arg在HPH大鼠中,通过影响ET-1的释放调节HPH。
AIM:To investigate effect of exogenous L-arginine(L-Arg) on release of endothelin in hypoxic pulmonary hypertension(HPH)rats.METHODS:Forty rats were randomly divided into four groups:control group,hypoxic group,hypoxia+L-NAME group and hypoxia+L-Arg group.Pulmonary artery mean pressure(mPAP),dry weights of right ventricle(RV),left ventricle+septum(LV+S)were measured and RV/(LV+S)ratio was calculated.Plasma endothelin-1 (ET-1) concentration was detected by using RIA.RESULTS:mPAP was significantly higher in rats of hypoxic group than that of control group( P <0 05) and it was much lower in rats of hypoxia+L-Arg group than that of either hypoxic group ( P <0 05) or hypoxia+L-NAME group( P <0 05).Weight of RV/(LV+S)ratio was increased in rats of hypoxic group compared with that of control group( P <0 01).While,it was increased in rats of hypoxia+L-NAME group compared with that of either hypoxic group( P <0 05)or hypoxia+L-Arg group( P <0 01).Plasma ET-1 level in rats of either hypoxic group or hypoxia+L-NAME group was higher than that of either control group or hypoxia+L-Arg group( P <0 01).mPAP positively correlated with plasma ET level( r =0 682, P <0 05) CONCLUSION:L-Arg played a modulatory role on the development of HPH through impacting the ET release from hypoxic rats.
